Raster Image Correlation Spectroscopy (RICS)

Instead of correlating the intensity over time, here one also correlates also over space by very rapidly scanning a single detection volume and thus capturing the diffusion of individual molecules. A spatial correlation analysis subsequently allows one to calculate the diffusion coefficient(s).

 

Examples of use:

  • Binding (dynamics) or colocalization of two fluorescently-labelled molecules separated (especially by more >10nm).
